Dear Editor,

Artemisinin (or Qinhaosu) is the active component of the wormwood plant Artemisia annua and has been used for centuries in traditional Chinese medicines to treat fever and malaria. With the emergence of drug-resistant strains artemisinin remains the most effective weapon against malaria today, but despite 30 years of effort, its mode of action is still a mystery. Previous studies have been restricted to superficial observation or testing putative cellular targets, rather than searching for targets in an unbiased experiment and in function-relevant manners.

We take advantage of a powerful yeast model to understand the action of this important drug at molecular level. Genetic approach to identifying relevant genes or pathways makes few assumptions about the kinds of molecules and mechanisms that regulate this process and, thus, is well positioned to uncover new molecules and mechanisms. Our results from this study provide crucial clues as to how artemisinin is activated and how the activated artemisinin damages the cell. We believe Nature is a superb choice for this ground-breaking discovery.
